Effects Chain being ignored?

I'm fairly new and using a registered version of Reaper.

Here's my current problem:

I have an effects chain with 3 effects: ReaGate, ReaEQ and ReaComp. My track (containing voice) has the FX enabled (little green light on the track, all 3 effects in the chain are ticked), yet they don't seem to be doing anything.

When I flick the effects chain on and off, there is no discernable difference in the audio. I drag the gate slider up, and nothing is cut out (even when I drag it to the top).

I'm mostly trying to remove the room noise, but it's just not happening. Do I need to re-install?
