There might probably also be other options, but one way trying to replicate the things shown in the video is basically:
1) Creating an
Aggregate Audio Device (as shown in the video) ...
2) ...
and selecting the Aggregate Audio Device then
as Audio Device in Reaper's Preferences
3)
Adding Audio Track #1 (= Ableton Live Track 3 in the video) and assigning an appropriate audio input (Stereo-Input 1/2 in the video) as Track Input:

Adding Audio Track #2 (= Ableton Live Track 4 in the video) and placing an audio sample on it
Then in the Track
I/O-Routing Panel ** of
Audio Track #2:
5) Selecting an appropriate audio output (output 5 in the video) as
Audio Hardware Output at the bottom left
6) Unchecking the
Master/Parent send checkbox at the top left
